Purification and characterization of pectin acetylesterase from orange peel

Abstract

Pectin acetylesterase was purified from orange peel using ammonium sulphate fractionation, followed by chromatography on Sephadex G-25 superfine, mono S and Superose 6. The purified enzyme has a Mr of 29 000. It has no pectin methylesterase activity, but rapidly de-esterifies triacetin. The pectin acetylesterase activity is increased 4.8-fold by prior incubation of sugarbeet pectin with pectin methylesterase, which suggests that the acetylesterase preferentially deacetylates galacturonyl residues containing a free C-6 carboxy group.
