This chapter describes methods for random transposon mutagenesis of pathogenic leptospires, identification of transposon insertion sites using direct sequencing from genomic DNA and a nested PCR utilizing degenerate oligonucleotides, and methods for testing mutant attenuation in the hamster model of infection.",
