Obesity is increasing at epidemic rates throughout the world and the brain is prone to excessive lipid
accumulation and mitochondrial dysfunction in obesity. This proposal addresses mitochondrial and lipid
metabolism in brain cells controlling food intake and body weight gain. We will create a unique genetic model
by deleting CPT1c, a mitochondrial enzyme involved in fat metabolism, in POMC brain cells that suppress
food intake. We will, 1) examine the effect on brain circuits regulating food intake, 2) the susceptibility to dietinduced
obesity and 3) glucose intolerance. We expect to identify for the first time novel mitochondrial
molecular targets in the brain to treat obesity and diabetes.
