Vancomycin and related antibiotics are vital compounds used to treat drug-resistant bacterial infections. These antibiotics are too complex to chemically synthesise and we are dependent on natural biosynthesis for their pharmaceutical production. The overall goal of our research is to overcome these limitations and design novel biosynthetic machineries to make new antibiotics with the potential to overcome ever increasing examples of bacterial antibiotic resistance. In this project, we propose to reconstitute the complete peptide synthesis machinery that makes vancomycin and related antibiotics – a non-ribosomal peptide synthetase – in order to learn how to reengineer such systems to produce new antibiotics.
