Abstract Title: Improved anaesthetic formulations comprising neuroactive steroids
The present invention relates generally to the field of drug delivery systems for neuroactive steroid anaesthetic agents. More particularly, anaesthetic and sedative formulations are provided in the form of host/guest preparations comprising one or more neuroactive steroid anaesthetics and a cyclodextrin. Particular cyclodextrins contemplated include sulfoalkyl ether cyclodextrins and modified forms thereof, especially sulfobutyl ether β-cyclodextrin. The neuroactive steroid may be alphaxolone, alphadolone, allopregnanolone or eltanolone (pregnanolone). The formulation may additionally comprise an antimicrobial agent, a preservative, a buffer and/or a copolymer, particularly hydroxypropyl methylcellulose (HPMC), polyvinyl pyrrolidone (PVP) or carboxymethylcellulose. The composition may be administered by infusion or bolus.
